Biological activity of NNMTi
NNMTi is a potent inhibitor of nicotinamide N-methyltransferase (NNMT) (IC50=1.2μM), 5-amino-1-methyl quinoline-1-ium iodide can selectively bind to the NNMT substrate-binding site Residues.
NNMTi can promote the differentiation of myoblasts in vitro and enhance the ability of fusion and regeneration of muscle stem cells in aged mice.
In vitro studies
NNMTi(10-30µM;96hours) produces a concentration-related increase in myoblast differentiation on C2C12 myoblast differentiation.30µMNNMTiresultsin18%MHC positive Chemicalbookemyotubenuclei, representing a 45% increase in the extent of myoblast differentiation compared to untreated differentiating myoblasts(12%MHC-positive myotube nuclei).
